Do we have to enable/activate the notificaton activity for the first time???
If yes then how do we do it? i have used this stage in the sequencer stage,
job runs but the mails are not sent.

No, there's nothing to enable/activate for this. Talk to your SysAdmins if you are having a problem with 'sendmail' on your system, as that is what DataStage uses.
