Change in dsenv file -

Need some info - if we change the dsenv file to add db2 instance - do we need to stop and start the datastage?
what are the steps need to take if any?

Yes. Search for the 'How to' part.
No special steps. Make a copy of the dsenv file just to be on the safe side. Make changes. Stop engine. Start engine.
Make sure no one is connected to the engine at that time. You dont want angry folks on your back
